溶藻弧菌HY9901 ahr基因克隆与序列分析(英文)

CLONING AND CHARACTERIZATION OF THE GENE(ahr) ENCODING ATP-DEPENDENT DNA HELICASE REP FROM VIBRIO ALGINOLYTICUS STRAIN HY9901

  • 摘要: Touchdown PCR扩增溶藻弧菌依赖ATP的DNA解旋酶rep基因部分序列,得一1.2kb片段,再以反向PCR和巢式PCR联合扩增其侧翼序列,拼接得一由2016 bp组成,共编码671 aa的完整基因。该基因演绎的氨基酸序列与几种弧菌的同源性都比较高,与副溶血弧菌RIMD 2210633、溶藻弧菌12G01、Vibriosp.Ex25、创伤弧菌YJ016、霍乱弧菌RC385、灿烂弧菌12B01、费氏弧菌ES114及V.angustumS14分别为96%、95%、95%、94%、92%、91%、89%、86%。

     

    Abstract: A112 kb internal fragment of the ATP-dependentDNA helicase rep gene from Vibrio alginolyticus strain HY990. was amplified by PCR with designed primers and the unknown flanking sequences of 5p-and 3p-ends of the ahrgene were characterized by using inverse PCR and nested PCR. Sequence analysis showed theahr gene contained a 2016 bpORF en-coding 671 amino acids. Compared with other relevantvibrios, the deduced amino acid sequence of the ATP -dependent DNA helicase rep precursor from Vibrio alginolyticus strain HY990. possesses very high similarity with them.
